When they compose and try to send, they may get a relaying error if they are using an alien ISP setting.
Solution is to change the settings in outlook temporarily to your outgoing server.

Ocassionally when in the office other clients computers connect to the network to get their mail, sometimes it works sometimes it doesnt. Should they change their pop3 and smtp servers to ours? Why does it just work fine in our network/ISP, and not with others? Is this an ISP restriction?

For the 550 - it looks as though it won't deliver to any addresses that are not local to it, i.e. it won't relay for you.
For your POP problem,.. I've had it before when a pop mailbox is sort of in a 'locked state' and won't allow connections until the process that is accessing it times out,.. after some time it gets released.

You will find that POP3 works for most ISP's but SMTP won't unless authorized on the right network.
I dial in on my laptop and am able to receive email, but cannot send it until I return home and send it once connected to my broadband ISP.
You could try authenticated SMTP relay, in the SMTP settings. this will only work if the ISP supports it.

I have just tried this and I can send out on a "foreign" computer, but not recieve anything at all. Have setup both POP3 and SMTP as our mail servers, as I said resolved the sending issues, but cannot recieve, keeps prompting for password even though I know it is correct.
Furthermore when I reply from an "office" pc to the "foreign" pc, I get an undeliverable email saying..........
